Baclosan is a muscle relaxant from a group of drugs that have an anti-inflammatory and analgesic effect. Active ingredient: Baclofen.

Centrally acting muscle relaxant; GABAB receptor agonist. By reducing the excitability of the terminal sections of afferent sensory fibers and suppressing interneurons, it inhibits mono- and polysynaptic transmission of nerve impulses; reduces pre-tension of muscle spindles.

Does not affect the transmission of impulses at neuromuscular synapses. For neurological diseases accompanied by spasticity of skeletal muscles, it weakens painful spasms and clonic convulsions.

Increases the range of motion in the joints, facilitates passive and active kinesitherapy (physical exercises, massage, manual therapy).

Available in tablets of 10 mg and 25 mg. Baclosan 10 mg tablet is marked with a dividing line. The 25 mg tablet has no dividing risks. The shape is round, biconvex, white.

Baklosan tablets composition

1 tablet contains:

Active substance: baclofen 10 mg or 25 mg. Excipients: lactose, potato starch, gelatin, talc, magnesium stearate, ethylcellulose.

Pharmacodynamics

A centrally acting muscle relaxant, a derivative of gamma-aminobutyric acid (GABAb-stimulator). By reducing the excitability of the terminal sections of afferent sensory fibers and suppressing interneurons, it inhibits mono- and polysynaptic transmission of nerve impulses; reduces pre-tension of muscle spindles. Has no effect on neuromuscular transmission. For neurological diseases accompanied by spasticity of skeletal muscles, it weakens painful spasms and clonic convulsions; increases the range of motion in the joints, facilitates passive and active kinesitherapy (physical exercises, massage, manual therapy).

Pharmacokinetics

Absorption is high. The maximum concentration in blood plasma is achieved 2-3 hours after taking the drug. Connection with blood plasma strips - 30%. Penetrates the placental barrier and is excreted in breast milk. Metabolized in the liver. Excreted by the kidneys (mainly unchanged).

Baklosan contraindications

Hypersensitivity, epilepsy, convulsions (including history), psychosis, Parkinson's disease, peptic ulcer of the stomach and duodenum, lactase deficiency, lactose intolerance, glucose-galactose malabsorption, pregnancy and lactation, children under 3 years of age.

Carefully

Cerebrovascular insufficiency, cerebral atherosclerosis, chronic renal failure, old age, children under 12 years of age.

Baklosan tablets dosage and method of administration

Inside, during meals. If you miss the next dose, you should not take a double dose of the drug.

Adults: Initial dose - 5 mg (1/2 tablet of 10 mg) 3 times a day, followed by increasing the dose every 3 days by 5 mg until a therapeutic effect occurs (usually up to 30-75 mg per day). The maximum daily dose is 100 mg. The final dose is set so that the decrease in muscle tone does not lead to excessive myasthenia gravis and does not impair motor function. For hypersensitivity, the initial daily dose is 5-10 mg, followed by a slower increase. For chronic renal failure and during hemodialysis - daily dose - 5 mg (1/2 tablet of 10 mg). In patients requiring high doses of bacpofen (from 75 mg to 100 mg per day), the drug should be used in 25 mg tablets. In patients over 65 years of age, the dose of the drug should be increased with caution, due to the increased risk of side effects.

Children: Initial dose - 5 mg (1/2 tablet of 10 mg) 3 times a day. If necessary, the dose can be increased with caution every 3 days by 5 mg until a therapeutic effect occurs. The following doses are usually recommended: For children from 3 to 6 years old, 120-30 mg per day, from 6 to 10 years old - 30-60 mg per day. In children over 10 years of age, the maximum daily dose is 2.5 mg/kg body weight; initial dose -1.5-2.0 mg/kg body weight. The drug is withdrawn gradually (over 1-2 weeks).

Interaction with other drugs

The attending physician must be aware of other medications the patient is taking in parallel.

Together with other drugs that affect the central nervous system and alcohol, the sedative effect of the active substance increases.

If lithium salts are present in another medicine, the patient will experience hyperkinetic symptoms.

Muscle tone decreases when taken together with antidepressants. When taking drugs to lower blood pressure in parallel, it is necessary to adjust the doses of both drugs.

Kidney failure can occur when taking Baclofen with Ibuprofen (Nurofen). When taken together with Levodopa, problems with clarity of thinking and consciousness are noted.

Baklosan side effects

Depending on the frequency of occurrence, the following groups of side effects are distinguished: very rare - less than 0.01%, rare - more than 0.01% and less than 0.1%, infrequent - more than 0.1% and less than 1%, frequent - more than 1 % and less than 10%, very common - more than 10%.

From the nervous system: very common - drowsiness, sedation; frequent dizziness, weakness, fatigue, confusion, gait disturbance, insomnia, euphoria, depression, asthenia, ataxia, tremor, hallucinations, nightmares, nystagmus, dry mouth; rare - paresthesia, dysarthria.

From the cardiovascular system: frequent - decreased cardiac output, arterial hypotension.

From the gastrointestinal tract: very common - nausea; frequent, vomiting, constipation, diarrhea, dyspeptic symptoms; rare - abdominal pain, dysgeusia, liver dysfunction.

From the respiratory system: frequent - respiratory depression.

From the musculoskeletal system: frequent - myalgia.

From the senses: frequent - accommodation paresis, visual disturbances.

From the skin and subcutaneous tissue: common - hyperhidrosis, rash.

From the kidneys and urinary system: frequent - polyuria, enuresis, dysuria; rare - urinary retention.

From the reproductive system: rare - erectile dysfunction.

General disorders: very rare - hypothermia.

In isolated cases, an increase in spasticity was observed, which was regarded as a paradoxical reaction to the drug.

Overdose

Symptoms: muscle hypotension, depression of the respiratory center, confusion, coma; After the return of consciousness, muscle hypotension may persist for 72 hours.

Treatment: plenty of fluids, diuretics; in case of respiratory depression - artificial ventilation. There is no specific antidote.

Interaction with other drugs

Enhances the effect of drugs that depress the central nervous system, antihypertensive and anti-gout drugs, ethanol. Tricyclic antidepressants increase the decrease in muscle tone; Levodopa, carbidopa in combination with levodopa increase the risk of hallucinations, confusion, and agitation.

Side effects are often transient in nature, they should be distinguished from the symptoms of the disease for which treatment is being carried out: with concomitant epilepsy, treatment is carried out without discontinuation of anti-epideptic drugs. In patients with liver disease and diabetes mellitus, it is necessary to periodically monitor the activity of liver transaminases, alkaline phosphatase, and blood glucose concentration. During treatment, you should refrain from potentially dangerous activities associated with the need for increased attention and rapid mental and motor reactions.
